蛇是爬行纲动物演化史中最迟出现的类群,最早的蛇化石出土于距今七千多万年的中生代白垩纪地层内。蛇和蜥蜴的亲缘关系密切,许多学者经过研究发现:较原始的细盲蛇科的蛇中,体鳞呈覆瓦状排列,腹鳞小而分化不明显;上颌骨、腭骨、翼骨等颅骨密接,方骨短,限制了蛇嘴的极度张开;下颌保留着冠状骨;有乌喙骨和腰带骨。

巨蜥科的分叉之舌,以及美洲鬣蜥科(Iguanidae)的椎骨结构和血红蛋白经蛋白酶水解所产生的肽类指纹图等,均与蛇相似。更有甚者的是蜥蜴目中不但具有四肢退化而呈蛇形的蛇蜥科、蚓蜥科的种类。

并且还能在不同类型的蜥蜴之间看到其肢体长度比例上逐渐缩短直至萎缩消失的过程。这些实例都无可争辩地证实了蛇是渊源于蜥蜴,并循着身体延长和丧失四肢演化而从蜥蜴主干上所分化出的一个分支。
